Mutation and Fermentation of Hypsizigus marmoreus

摘要: 采用紫外线诱变技术对真姬菇进行诱变育种,通过初筛、复筛,获得6号菌株为优选菌株。该菌株摇瓶发酵菌丝体干重达22.700 mg•mL-1,胞内多糖为4.0634 mg•mL-1,胞外多糖为1.7220 mg•mL-1,总糖产量达5.7834 mg•mL-1,比其他各菌株产量都高。对6号优选菌株进行碳氮源等培养条件优化筛选,得出其最适碳氮源及浓度为:可溶性淀粉2.0%,麦芽糖3.0%,牛肉膏0.6%,蛋白胨0.2%;在该条件下真姬菇6号菌株的菌丝干重为20.2580 mg•mL-1,胞内多糖产量为4.2587 mg•mL-1,胞外多糖产量为2.234 2mg•mL-1。Abstract: By means of UV radiation mutagenesis on Hypsizigus marmoreus,the mutation breeding strain No.6 was resulted after repeated screenings.Mycelia of the fermentation strain had a dry weight of 22.700 mg•mL-1.It contained 4.063 4 mg•mL-1 intracellular polysaccharides,1.7220 mg•mL-1 exopolysaccharide and 5.783 4 mg•mL-1 total sugars.The yield was higher than that from other strains.The optimized culture.composition was found to consist of 2.0% soluble starch,3.0% maltose,0.6% beef extract and 0.2% peptone.Cultivated with the optimized formulation,No.6 H.marmoreus mycelium had a dry weight of 20.2580 mg•mL-1.Its intracellular polysaccharide content increased to 4.2587 mg•mL-1 and exopolysaccharide to 2.2342 mg•mL-1.
