Preparation and biological characteristics of neutralizing monoclonal antibodies against newcastle disease virus

摘要: 应用杂交瘤技术,以纯化的NDV-lasota为免疫原结合间接ELISA筛选到了24株抗NDV单抗,其中3 株(McAb-14、McAb-18和McAb-21)具有中和活性,并与其血凝抑制效价具有很大的相关性。McAb-18和 McAb-21的免疫球蛋白亚类都属于IgG2a;McAb-14为IgG2b。特异性鉴定结果表明:3株抗NDV的单抗均不与马立克病毒、传染性支气管炎病毒、传染性法氏囊炎病毒、减蛋综合征病毒、传染性喉气管炎病毒等发生交叉反应。用这3株中和单抗的任2种混合液对鸡进行了被动免疫治疗和免疫预防试验,结果表明:McAb-14+McAb-18组合的免疫预防和治疗效果最好。Abstract: Twenty-four monoclonal antibodies (McAbs) against newcastle disease virus (NDV) were obtained by using hybridoma technique.Among them, McAb-14, McAb-18 and McAb-21 had strong neutralization characteristics, and there was strong correspondence between the neutralization efficacity and the haemagglutination inhibition value.The McAb-18 and McAb-21 belonged to IgG2aclass, and McAb-14 was TgG2bclass.No crossreaction was detected between the McAbs and infectious bronchitis virus, Marek’s disease virus, infectious bursal disease virus, egg drop syndrome virus and infectious laryngotracheitis virus.Two of the three neutralizing McAbs were equivalent mixed randomly; the passive immunization treatment and immune preventive test of the mixture were examined in chicken.The results showed that McAb-14+McAb-18 combination was the most effective.
