Prokaryotic Expression of the NS1 Gene for Avian Tembusu Virus

摘要: 根据GenBank中登录的禽坦布苏病毒 (avian Tembusu virus, ATMV) WR株非结构蛋白NS1基因设计特异性引物, 采用RT-PCR方法从禽坦布苏病毒WR株核酸中扩增其NS1基因片段, 并将其克隆到原核表达载体pGEX-4T-1上构建原核表达重组质粒pGEX4T-NS1, 进行条件优化诱导表达, 将表达的目的蛋白经SDSPAGE分析。结果表明, 分子量约为62kD的重组目的蛋白得到表达。Abstract: A pair of primers were designed and synthesized according to the nanostructure protein (NS1) gene of avian Tenbusu virus WR strain available in GenBank.The NS1gene was amplified by RT-PCR and cloned into the prokaryotic expression vector pGEX-4T-1, then induced into E.coli with IPTG.The SDS-PAGE analysis of expressed protein demonstrated that the recombinant NS1protein of 62kD had got a high expression, which laid a good foundation for developing in molecular diagnosis and the function of the NS1gene for avian Tenbusu virus.
