Orthogonal Optimization for Propagation of Clustered OncidiumBuds

摘要: 以文心兰‘小樱桃’花梗腋芽诱导出的丛生芽为增殖培养材料, 采用正交设计法, 研究基本培养基、6-BA、NAA和水解乳蛋白 (LH) 4种因素对文心兰丛生芽增殖的影响, 以期筛选出各因子的最佳水平, 建立文心兰‘小樱桃’丛生芽增殖培养体系。结果表明:各试验因素对文心兰丛生芽增殖影响的主次关系为6-BA>基本培养基>NAA>LH;筛选出文心兰丛生芽增殖的最佳培养基配方为改良1号+6-BA 3.0mg·L-1+NAA0.1mg·L-1+LH 0.5g·L-1, 45d平均增殖系数达6.53, 有效提高了繁殖效率, 为其种苗工程化育苗提供了技术基础。Abstract: Peduncle axillary buds of the Little Cherry, Oncidium, were induced as explants.The orthogonal design was used to evaluate the effects of the culture medium containing inorganic substances, 6-BA, NAA and LH on the propagation of Oncidiumbuds.The result showed that 6-BA, the base medium, NAA and LH affected significantly on the bud multiplication.The optimal medium was the Improved Formulation No.1, which consisted of the base medium, 6-BA 3.0mg·L-1, NAA 0.1mg·L-1 and LH0.5g·L-1, and the average propagation coefficient was 6.53 in 45days.The breeding efficiency was effectively improved.The result appeared to provide a basis for the commercialization of the seedling propagation.
