DNA Fingerprinting by ISSR Markers for 17 Blueberries

摘要: 利用ISSR分子标记技术分析17个蓝莓品种的遗传多样性, 并构建DNA指纹图谱。从60个ISSR引物中筛选出10个稳定的多态性引物。采用NTSYS-PC 2.0软件进行聚类分析, 17个蓝莓品种的遗传相似系数为0.660.86, 在0.69的水平可分为4个品种群。引物UBC825、UBC862可将17个蓝莓品种区分开, 并依此建立了数字化的DNA指纹图谱, 为蓝莓品种分类、快速鉴定提供依据。Abstract: Inter-simple sequence repeat (ISSR) analysis was applied for the determination of the genetic diversity, as well as the establishment of the DNA fingerprints, for 17 selected blueberry cultivars.Ten out of 60 ISSR primers were chosen for the study.Cluster analysis revealed that the genetic similarity coefficients of the 17 samples ranged from 0.66 to 0.86.Using the coefficient threshold of 0.69, 4classifications were formed for the blueberries.They could be positively distinguished by 2ISSR primers (i.e., UBC825 and UBC862) , and used to obtain the digital DNA fingerprints.
