Breeding of Bacillus licheniformis Mutant by ARTP and Optimization of Bacitracin-producing Process

摘要: 为获得高产杆菌肽菌株,对海水养殖环境中分离的产杆菌肽菌株地衣芽孢杆菌ZZ061进行ATRP诱变,以藤黄微球菌为指示菌,采用抑菌圈法进行筛选,获得一株产杆菌肽水平明显提高的诱变株SF1406,其摇瓶效价比野生菌株提高130%,达到560U·mL-1。采用正交试验优化SF1406产杆菌肽培养基,结果表明:在玉米淀粉含量为3%,硫酸铵含量为1.5%,豆饼粉含量为1%,蔗糖含量为3%时效果最佳,杆菌肽效价进一步提高44%,达到810U·mL-1。Abstract: The bacitracin-producing Bacillus licheniformis ZZ061 was isolated from an aquaculture facility.From the isolated bacteria,mutants with enhanced capability of generating bacitrcin were bred by means of ARTP mutagenesis.In the process,Micrococcus luteus was used as the indicator bacterium,and the bacteriostatic circle method employed for the screening.One of the mutants,SF1406,showed the greatest increase,i.e.,130% higher than the wild strain,in its bacitrcin productivity.Subsequently,an orthogonal experiment was conducted to optimize the culture medium formulation.It was found that the medium consisting of 3% corn starch,1.5%ammonium sulfate,1%soybean powder and 3%sucrose rendered the most desirable results with an additional 44%productivity increase on the yield of 810U·mL-1.
