龙眼新品种(品系)的ISSR指纹图谱研究初探

DNA Fingerprinting of 17 Dimocarpus longan Germplasms by Using ISSR Markers

  • 摘要: 利用ISSR分子标记技术,以冬宝9号、高宝等6份龙眼新品种(品系)与我国11份主栽品种为试材,构建17份龙眼品种(品系)的DNA指纹图谱。从30条ISSR引物中筛选出5条稳定的多态性引物。采用NTSYS-PC 2.0进行聚类分析,17份龙眼种质的遗传相似系数为0.75~0.93,在0.785的水平可将试材分为4个类群。引物py-9可将17个龙眼品种(品系)区分开,并依此建立了数字化的DNA指纹图谱,为探索龙眼新品种的鉴别、资源评价利用等方面提供了重要的依据。

     

    Abstract: The inter-simple sequence repeat(ISSR) method was applied to assess the genetic makeup of 17,including 6 new longan varieties.Out of the 30 ISSR primers,5 were selected for the analysis.Cluster analysis showed that the genetic similarity coefficients ranged from 0.75 to 0.93,and that the 17 cultivars could be clustered into 4 groups using a genetic similarity coefficient threshold of 0.785.These varieties could be satisfactorily distinguished by a single ISSR primer,py-9,and established their digital DNA fingerprints accordingly.The results provided an important basis for identifying new longan varieties,as well as evaluating and utilizing the germplasm resources.
