水稻类病斑及早衰突变体lms1的鉴定及基因初步定位

Identification and Gene Mapping of a Lesion Mimic and Senescence Mutant lms1 in Rice

  • 摘要: 通过筛选籼稻恢复系明恢86的组培变异后代,获得1个类病斑及早衰突变体lms1(lesion mimic and senescence 1)。lms1植株生长至拔节期开始在叶片上出现黄褐色小斑点,随后斑点逐渐扩展至大部分叶片和茎组织;生长至抽穗期后呈现早衰,穗、茎、叶明显干枯,并快速衰亡。RT-PCR分析表明,在呈现类病斑性状叶片组织的lsm1中,病程相关基因PBZ1、PAL1表达明显高于其在野生型叶片组织中的水平。遗传分析表明,lms1的突变性状受单隐性核基因控制。利用9311与lms1配置的F2及F3群体进行基因定位,将lms1基因定位在水稻第11染色体长臂末端。

     

    Abstract: A lesion mimic and senescence mutant,designated as lms1,was isolated from the tissue culture progeny of indica rice restorer line Minghui-86.lms1plants started to appear small brown spots on the leaves in elongation stage,and the spots gradually spreaded to whole leaves and sheaths.With the growth of the plants to heading stage,lms1displayed a senescent phenotype,whereas leaves,stems,and panicles dried obviously,resulting in whole plants′death.RT-PCR analysis revealed that the expression of pathogenesis-related genes PBZ1and PAL1 was induced in lms1leaf tissues.Genetic analysis suggested that the loci for the mutant phenotype was controlled by a single recessive gene.F2and F3 populations derived from a cross between 9311and lms1 were used for gene mapping,and the lms1locus was mapped close to the terminal region of the long arm of chromosome 11.
