中国水仙类黄酮3-氧-葡糖基转移酶基因的克隆与序列分析

Cloning and Expression Analysis of Flavonoid 3-O-glucosyltransferase Gene from Narcissus tazetta.var.Chinensis

  • 摘要: 采用RT-PCR和RACE技术,从中国水仙上分离克隆得到1个葡糖基转移酶基因,命名为Nt3GT,其cDNA全长1 682bp,包含有1个1 461bp完整阅读框架(ORF),编码487个氨基酸,具有Glycostranf1superfamily蛋白保守区。系统进化分析表明,中国水仙与马铃薯属的3GT亲缘关系最近。半定量RT-PCR结果显示,Nt3GT在叶片和花中均有表达,各个时期的叶片中表达量稳定,但花中的表达量不稳定,在抽葶期花苞未检测到表达,在盛花期的表达最高。

     

    Abstract: The Flavonoid 3-O-glucosyltransferase gene was cloned from Narcissus tazetta.var.Chinensis by RACE and RT-PCR.The full lenght of Nt3 GT was 1 682 bp with an ORF of 1461 bp,which encoded a protein of487 amino acids with a conservative domain of Glycostranf1superfamily.Phylogenetic analysis demonstrated that Nt3 GT was in the closest relationship with Solanum tuberosum(AAZ76827.1),Solanum tuberosum(AAX63403.1)and Solanum pinnatisectum(AAY88192.1).Semi-quantitative RT-PCR revealed that Nt3 GT could expressed in leaves and flower,and the expression in leaves was stable in different times.However,the expression in buds was unstable.It could not be detected during scape-sprouting,and was the highest in flowering.
