花叶金线莲茎段诱导植株再生培养技术研究

Induction Plant Regeneration with Stem Segments of Anoectochilus roxburghii (wall.) lindl. Huaye

  • 摘要: 以花叶金线莲茎段为外植体,采用正交设计,探讨基本培养基(MS、B5、改良MS)、植物生长调节剂(TDZ、6-BA、KT、IBA)等对其茎段诱导、增殖及生根等关键环节的影响,以期建立花叶金线莲茎段诱导植株再生培养技术。结果表明:各试验因素对花叶金线莲茎段诱导丛生芽影响的主次关系为基本培养基>KT>6-BA>TDZ;筛选出适宜的增殖培养基配方为改良MS+6-BA 2.0 mg·L-1+KT 0.5 mg·L-1+NAA 0.1 mg·L-1+白糖30.0 g·L-1+琼脂粉3.0 g·L-1+卡拉胶3.0 g·L-1,60 d平均增殖系数达4.58;筛选出适宜生根的培养基配方为改良MS+IBA 0.3 mg·L-1+活性炭0.5 g·L-1+白糖20 g·L-1+琼脂粉3.6 g·L-1+卡拉胶3.6 g·L-1,培养60 d,生根率为100.0%。

     

    Abstract: Stem segments of Anoectochilus roxburghii (wall.) lindl. Huaye were used as the explants to induce bud clumps. Varied medium (i.e., MS, B5 or improved MS) and phytohormone (i.e., TDZ, 6-BA, KT or IBA) were applied for the tissue regeneration experiment of an orthogonal design. It was found the major factors affecting the bud multiplication to be basal medium, KT, 6-BA and TDZ. The optimal multiplication was achieved by using the improved MS+6-BA with additions of 2.0 mg·L-1+KT 0.5 mg·L-1+NAA 0.1 mg·L-1+sugar 30.0 g·L-1+agar powder 3.0 g·L-1, and carrageenan 3.6 g·L-1. An averaged propagation coefficient of 4.58 in 60 days was obtained. For rooting, the improved MS+6-BA with added 0.3 mg ·L-1+AC 0.5 g·L-1+sugar 20.0 g·L-1+agar powder 3.6 g·L-1 and carrageenan 3.6 g·L-1, rendered a 100.0% rate in 60 days.
