南方根结线虫生防菌JXJA01鉴定及其杀线活性研究

Identification and Nematicidal Activity against Meloidogyne incognita of Actinomyces

    目的 对南方根结线虫生防菌JXJA01进行鉴定并探究其杀线活性,以拓展根结线虫病的生防资源。
    方法 以本实验室前期从赣州脚板薯根际土壤中分离到的1株放线菌菌株JXJA01为研究对象,综合形态学特征、生理生化特性和分子进化分析结果进行种属确定,并将菌株JXJA01发酵上清液分别与线虫悬液和新鲜卵囊共同培养进一步验证其杀线活性。
    结果 确定该菌株为欧洲疮痂链霉菌(Streptomyces europaeiscabiei)。不同稀释倍数菌株JXJA01发酵上清液中以2倍稀释液对根结线虫二龄幼虫(J2s)的生物活性最强, 24 h时校正死亡率为95.53%;对根结线虫卵孵化也具有较强的抑制作用,处理7 d时的孵化抑制率达44.00%;其对盆栽番茄根结线虫病同样具有较好的防效,60 d时的防效达40.88%,还可显著增加番茄的根鲜重,增幅较对照高71.42%;此外,该菌株发酵上清液对温度和光照不敏感,4 ℃条件下储藏21 d不会降低杀线虫活性。
    结论 该菌株可作为根结线虫的生防资源,用于后续杀线生防菌剂的研发。

     

    Abstract:
    Objective Identification and nematicidal activity against Meloidogyne incognita of an Actinomyces strain were investigated for root-knot disease control.
    Method An Actinomycete strain, JXJA01, previously isolated from the rhizosphere soil of Dioscorea alata Lirm. sp in Ganzhou, China was positively identified by morphological, physiological, and biochemical examinations as well as a molecular evolution analysis. The supernatant of JXJA01 fermentation broth was co-cultured with a suspension of the second stage parasite juveniles (J2) or fresh nematode eggs to determine the nematicidal activity of the isolated strain.
    Result JXJA01 was identified as Streptomyces europaeiscabiei. The 2-fold dilution of the supernatant of the bacterial fermentation broth displayed the strongest biological activity against J2s among all preparations with an adjusted mortality rate in 24h of 95.53%. It also had a robust suppressive effect on egg-hatching with a relative inhibition rate of 44.00% after 7 d of treatment. On potted tomatoes, it showed 40.88% efficacy in controlling the root-knot nematodes in 60d and significantly increased the fresh weight of roots by 71.42 % over control. Moreover, the fermentation supernatant was not sensitive to temperature or light and maintained the nematicidal activity in storage at 4 ℃ for 21 d.
    Conclusion Identified as S. europaeiscabiei, JXJA01 exhibited a promising potential to be used as a biological agent for controlling root-knot nematodes on tomatoes.
