Studies on Screening Somatic Cell Lines Resistant to Scab via Explant Culture of Wheat

  • 摘要: 以小麦幼穗为外植体,在含赤霉菌粗毒素的培养基上进行离体培养,研究筛选抗病细胞无性系的可能性及影响因素。结果表明,品种抗性不同,其愈伤组织的诱导和绿苗分化效果也不同。外植体以发育至1~3cm长的幼穗切段为宜。赤霉菌粗毒素浓度以10%(V/V)效果较好。从感病品种组培后代中筛选出6个抗病株系。
    Abstract: The possibility and affecting factors for screening the wheat somatic cell lines resistant to scab were studied by using explant culture of young spike on the medium containing crude toxin of F.graminearum The result showed that varieties with different resistance were different in callus induction and green plantlet differentiation; young spikes with 1-3 cm length were suitable for in vitro cultivation; the concentration of 10%(V/V) crude toxin was optimum one in the screening medium; six lines resistant to scab were obtained from scab sensitive varieties.
