Abstract:
Objective The pathogen of a newly emerging parasitic disease on Polygonatum cyrtonema Hua in Fujian was identified.
Methods A field investigation was conducted in P. cyrtonema production areas of Guangze County, Nanping City, Fujian to collect root and rhizosphere soil samples for isolating and identifying the parasitic nematodes. Based on morphological characteristics and molecular biological analysis, the isolates were screened, and the causal nematode was identified, with confirmation by an indoor infectivity test.
Results In the field, the nematode-infected P. cyrtonema plants grew slowly aboveground, with stunted growth and chlorotic leaves. The parasite was morphologically and molecular biologically identified as Rotylenchulus reniformis. In severely infested soil, the nematode population could be as dense as 2 300 nematodes per 100 mL. Mature females and gelatinous egg masses were observed on the roots of plants grown indoors in soil inoculated with R. reniformis for 60d, indicating completion of a life cycle of the parasites.
Conclusion The occurrence and causal agent of reniform nematode disease on P. cyrtonema were definitively identified in Fujian for the first time.
