反相HPLC法同时测定青梅中的7种有机酸

Simultaneous determination of seven organic acids in Prunus mume with reverse-phase high performance liquid chromatography

  • 摘要: 建立了用反相高效液相色谱法同时快速测定青梅中草酸、酒石酸、苹果酸、乳酸、乙酸、柠檬酸及琥珀酸等7种有机酸的方法.采用的色谱柱为Hypersil ODS C18(250 mm×.6 mm i.d.,5 μm),流动相为0.01 mol·L-1 KH2PO4(pH 2.8),流速为0.8 mL·min-1,柱温21℃,检测波长215 nm.有机酸的回收率为75.%~121.0%;相对标准偏差为0.78%~2.12%;检出限为0.022~1.290 μg·mL-1.测定了种不同青梅样品中有机酸的含量.

     

    Abstract: A method for simultaneous and rapid determination of 7 organic acids(i.e.,oxalic,tartaric,malic,lactic,acetic,citric and succinic acid)in Prunus mume with reverse-phase high performance liquid chromatography(HPLC)was developed.HPLC was performed on a Hypersil ODS C18 column(250mm×4.6mm i.d.,5 μm)at 21℃,using KH2PO4(0.01 mol·L-1;pH 2.8)as mobile phase at the flow rate of 0.8 mL·min-1 and the detection wavelength of 215 nm.The recoveries of the organic acids ranged from 75.4% to 121.0% with relative standard deviations of 0.78%-2.12% and the limits of detection 0.022-1.290 μg·mL-1.The results of the 7 organic acids in 4 P. mume samples are presented.
