Rapid determination of γ-amino butytic acid using amino acid analyzer

  • 摘要: 用氨基酸自动分析仪30 min短程序快速测定植物产品中的γ-氨基丁酸,并与53 min标准程序的测定值进行比较,试验表明两种程序测定结果一致。用γ-氨基丁酸标准品作精密度和回收率试验,结果表明:峰位置重现性变异系数为0.17%,峰面积重现性变异系数为0.23%,回收率为97.89%~101.2%。采用两种程序测定糙米和中药材中的γ-氨基丁酸,测定结果的相对误差小于3%。该改良方法稳定、快速、准确,适于γ-氨基丁酸的测定。
    Abstract: A method for rapid separating and determining γ-amino butytic acid(GABA) in plant products by using the HITACHI L-8800 amino acid analyzer was developed.The time required for the new procedure was 30 min in comparison to 53 min for the standard analysis.The determination results using the two methods were similar.At GABA concentration of 100 nmol·ml-1,the coefficient of variation of the peak potential and peak area was 0.17% and 0.23%,respectively.The recovery rates of GABA with the new procedures was 97.89%-101.2%.The relative errors of the determination in brown rice and traditional Chinese medicines by using the two methods were less than 3%.It was thus concluded that this modified method was stable,rapid,accurate and suitable for the determination of γ-amino butytic acid.
