两株菌草根际促生真菌筛选及其促生性能分析

Identification and Performance of Two Growth-promoting Rhizosphere Fungal Strains

    目的 本研究旨在挖掘具有优良促生特性的菌草根际微生物资源。
    方法 通过稀释平板法从菌草根际土壤中分离筛选出两株具有固氮能力潜力的真菌,并对其生理生化特性、ITS序列进行分子生物学鉴定及盆栽生菜促生效果进行研究。
    结果 结合形态学特征与ITS序列分析,鉴定结果显示Gt-3为黑曲霉(Aspergillus niger),Gt-29为长枝木霉(Trichoderma longibrachiatum)。生理生化特性试验结果表明,两株菌均具有解钾、产生铁载体、蛋白酶、淀粉水解酶、纤维素酶以及降解硅酸盐的能力。其中,菌株Gt-3还表现出溶磷特性。生菜盆栽促生试验结果显示,两株真菌对生菜生长均有显著促进作用。与对照组(CK)相比,Gt-29处理组的地上部鲜重增加了363.5%,株高增加了29.0%;Gt-3处理组的地上部鲜重增加了187.2%,株高增加了24.2%。此外,菌株处理后,生菜的营养品质也得到了显著提升(如可溶性糖、蛋白质、维生素C含量)及抗氧化酶活性(如POD、SOD、PPO和CAT酶活性)。
    结论 本研究成功从菌草根际筛选出两株具有固氮能力的多功能真菌Gt-3(黑曲霉)和Gt-29(长枝木霉)。两菌株均表现出产生铁载体、胞外酶及降解硅酸盐等多种促生特性,其中Gt-3还具备溶磷功能。盆栽试验结果证实,两菌株均能显著促进生菜生长、提升营养品质与抗氧化酶活性,为开发促生型微生物菌剂提供了优良的菌种资源与理论依据。

     

    Abstract:
    Objective Two strains of plant growth-promoting fungi were isolated from Juncao rhizosphere with performance determined in laboratory.
    Methods Fungi with high nitrogen-fixing capability for growth of plants were isolated from Juncao rhizosphere soil using the dilution plate method. Physiochemical characteristics and ITS sequences of two highly active strains were used to identify the species. A pot experiment using lettuce was conducted to evaluate the plant growth-promoting effect of the isolated strains.
    Results The two strains, Gt-3 and Gt-29, were identified to be Aspergillus niger and Trichoderma longibrachiatum, respectively. They could solubilize potassium, degrade silicates, and produce siderophores, protease, amylase, and cellulase. In addition, Gt-3 also exhibited a phosphate-solubilizing capability. The pot experimentation verified the ability of both strains in promoting the growth of lettuce. The presence of Gt-29 resulted in the increases of 363.5% on weight of fresh lettuce shoots and 29.0% on plant height, while Gt-3 187.2% on shoot weight and 24.2% on plant height over control. Furthermore, the nutritional quality (e.g., soluble sugar, protein, and vitamin C contents) and antioxidant enzyme activities (e.g., POD, SOD, PPO, and CAT) of the lettuce were significantly improved as well.
    Conclusion A. niger and T. longibrachiatum isolated from Juncao rhizosphere could promote, in different degrees, the growth, nutritional quality, and antioxidant enzyme activities of the potted lettuce. The two strains varied in siderophore production, extracellular enzyme secretion, and silicate degradation, and A. niger had an additional function of solubilizing phosphate.
