短葶山麦冬茎尖培养快速繁殖技术研究

Micropropagation for Liriope muscari (Desne.) Bailey

  • 摘要: 以短葶山麦冬茎尖为外植体,通过对其采取不同消毒时间、茎尖诱导分化、壮苗与生根及移栽条件等快速繁殖条件进行研究。结果表明:0.1%HgCl2消毒时间为12 min;短葶山麦冬茎尖分化培养基MS+BA 2.0 mgL-1 + NAA 0.2 mgL-1,茎尖分化率高,最高分化率达到90%。丛生芽增殖培养基选择MS+BA4.0 mgL-1 +NAA0.2 mgL-1为佳,一定高浓度的BA对麦冬茎尖丛生芽分化起着促进作用。短葶山麦冬壮苗生根培养基选择1/2MS+IBA0.5 mgL-1时最佳,生根系数达到6.87,泥炭土为最佳的移栽基质。
    Abstract: Shoot tips from Liriope muscari (Desne.) Bailey were used as explants to grow on media with varied sterilization time,under induced differentiation,as well as different strengthening and rooting treatments, and transplantation conditions. The shoot tips were cut from the plants and cultured on MS medium containing 2.0 mgL-1 BA and 0.2 mgL-1 NAA. The highest differentiating rate of 90% was achieved. The medium with 4.0 mgL-1BA and 0.2 mgL-1 NAA was suitable for multiple-shoot proliferation. A certain concentrations of BA promoted the shoot differentiation. The optimized medium for seeding and rooting was when MS was mixed with 0.5 mgL-1 IBA, which improved the root coefficient to 6.87. Peat soil was found to be the best medium material for the transplantation.
