Abstract
To alleviate a seed shortage on Pholidota cantonensis Rolfe, artificial culturing with medium optimization was investigated by using the plant tissue culture technology.The results showed that MS+20g·L-1 sucrose+8g·L-1 agar was an appropriate medium that allowed a germination rate above 70%.MS+50g·L-1 potato+20g·L-1 sucrose+8g·L-1 agar was desirable for the plantlet′s protocorm differentiation.Low concentrations of 6-BA and NAA in medium was better for bud protocorm proliferation and differentiation.The medium formulated with 1/2MS+0.5mg·L-1 NAA+20g·L-1 sucrose+8g·L-1 agar was optimal for rooting, as a rate of 100%was achieved.By using the combination of the above-mentioned media at respective stages, the plantlets of P.cantonensis Rolfe could be rapidly obtained from seeds.
